DETERMINATION OF POLYSACCHARIDE CONTENT IN TYPHOID VACCINE BY ELECTROPHORESIS METHOD USING 0.1M TRIS BUFFER

The rocket column electrophoresis method is one of the methods to quantify Vi Polysaccharide content in Vi typhoid vaccine, is the official method as recommended by WHO, and is included in Vietnam Pharmacopoeia V. However, Due to WHO recommendations, the National Institute for Control of Vaccines and Biologicals and manufacturers need to replace Bacbitan buffer solution with other buffer solutions because Bacbitan is a psychotropic chemical. The study to evaluate the suitability of using 0.1M Tris buffer solution in electrophoresis to determine Vi Polysaccharide content was conducted from November 2022 to April 2023 at the Department of Bacterial Vaccine Quality Control, National Institute for Control of Vaccines and Biologicals.
The results of evaluating the compliance with the accuracy criteria include repeatability CV=1.02% and intermediate accuracy CV= 2.02%, meeting the requirements according to the CV≤ 20% standard. From the research results, it has been proven that 0.1M Tris buffer solution can be used in electrophoretic testing to determine Vi Polysaccharide content in the Typhoid Vi vaccine at NICVB
