Identification of Vero 76 cell bank across generations by PCR sequencing the target genes psi beta and delta globin of Cercopithecus aethiops

Background/Purpose: Vero 76 Master Cell Bank and Vero 76 Working Cell bank are
specifically identified by used the psi beta and delta globin target gene sequencing method
of the vero 76 African green monkey kidney (Cercopithecus aethiops) cell line to identify
and evaluate the genetic stability of the Vero 76 Master Cell Bank (MCB) and Working
cell bank (WCB) were cultured several times on culture medium.
Methods: Sequencing target psi beta and delta globin genes
Results: The results of sequencing and comparison to determine the difference between
the cell samples in the transfected generations compared with the original Vero cell line
reference on the Genbank database (NCBI). The evaluation results showed that the cell
lines in the transfected generations did not differ from each other and compared with the
vero cell line reference on the Genbank database (NCBI), this confirmed the genetic
stability of the cell lines during the process establish and transfer the Vero 76 bank of
MCB and WCB.
Conclusion: We have successfully engineered a primer specifically for the amplification and
sequencing of the psi beta and delta globin genes of the African green monkey kidney
(Cercopithecus aethiops) in the Vero 76 cell line banks of (MCB and WCB) across generations.
Results from sequence analysis between generations are identical, proving genetic stability of
this cell line after multiple passages. Sequencing psi beta and delta globin genes specific to
African green monkey are also suitable to identify Vero76 cell line.
